Keep My Enemies, Loser is a point-and-click adventure game. The player moves Dangeresque through Perducci's restaurant, and can interact with people and items by choosing to look at, manipulate, or talk to them. Dangeresque narrates the actions he takes, for the benefit of the player. Puzzles frequently require completing several steps in sequence or combining multiple items.

Dangeresque must protect Perducci from four assassins, then make his escape from the mobster.
- Prevent Sultry Buttons from throwing a knife at Perducci.
- Prevent Unknown Assassin 1 from blowing a poison dart at Perducci.
- Prevent Unknown Assassin 2 from blasting Perducci with a laser-scope sniper rifle.
- Prevent Professor Experimento from dropping a satellite on Perducci.
- Mix a cocktail for Perducci with an extra ingredient: poison!

- Try to Get Killingyouguy.
- Hold up Killingyouguy with the plastic banana.
- Give Perducci the "all clear" before dealing with the current assassination attempt.
- Open the door to the kitchen before properly shielding Perducci.
- Walk into the laser sight while the sniper is aiming.
- Give the waiter orders in an incorrect arrangement.
- Answer the ringing phone.
- Be holding the phone when giving Perducci the "all clear".
- Give the cocktail a name that Perducci and/or Killingyouguy does not like.
- Give Perducci the cocktail without putting the poison dart in it.

- A "made man" is a mafia term for someone who has officially joined the ranks of the mafia, as opposed to being an associate.

- The title card first appears with the text "Keep My Enemies Closer", with a bullet hole removing the "C" as a comma is added to reveal the true title.
- Rif Simken is said to have 16 full hitters, 12 resists, and 37 MRI's.
- A no-hitter is a game where a pitcher plays all innings without a single batter on the opposing team getting a hit.
- A resist, like the full hitter and the MRI, is not a valid baseball statistic. The correct term is assist which, although a valid baseball statistic, is more generally associated with other sports such as soccer, basketball, and ice hockey.
- An MRI refers to Magnetic Resonance Imaging, which is used in brain scans. An RBI (Runs Batted In) is how many players scored runs while the player was at bat.

- Much like in the chair, a supposedly inanimate object based on Homestar Runner speaks and is brushed over.

- Dangeresque also watches The Facepaint Connection, one of Strong Bad's wrestling personas from yes, wrestling.
- Killingyouguy's favorite TV program is Doug the Dino, which was created by his actor, Strong Mad, and featured in the facts.
- Talking to the kitchen door will cause Strong Bad to order a waffle using the same intonation as Main Page 5.
- The WaiterID is ILKO418, referring to The Cheat's fake ID, Ilko Skevüld, and his birthday, April 18, in cheatday.
- "Dangeresque" taking a blow from Experimento's satellite looks identical to a similar stunt involving the Heavy Lourde in Dangeresque 2: This Time, It's Not Dangeresque 1.
- Killingyouguy uses a banana as a phone.
- If Dangeresque puts the poison dart in the Armzipan statue, Killingyouguy will overhear, but realize he doesn't have ears.
- In the epilogue, Dangeresque Too remarks that Rif Simken was a "homestar runner" for the Clarks, a reference to the origin of the body of work's name, where The Brothers Chaps' friend James Huggins described Mark Lemke as the "homestar runner for the Braves".

- Dangeresque puts a shield in Armzipan's hand to protect her from being turned to stone, a reference to the Ancient Greek myth of Perseus and Medusa.
