Most of the characters of the Homestar Runner universe are very different in physical characteristics from humans. However, there have been a few characters that more resemble humans. Notably, none of these characters have been fully visible in the main animation style.

- Arrow'd Guy — The tormentor of the Teen Girl Squad.
- Balding Man — A stereotype of the average middle-aged man.
- Crack Stuntman — The voice of Gunhaver of the Cheat Commandos.
- Crack Stuntman's Girlfriend — The voice of the New Bad Guy Girl Character on Cheat Commandos.
- A. Chimendez — Writer of Cheat Commandos and other TV shows.
- Limozeen — A stereotypical hair metal band, animated in best thing.
- Teeg Dougland — The manager of Limozeen.
- Advantage — An all-girl metal band, animated in The Limozeen Advantage.
- sloshy — A stereotypical alternative rock band, animated in Limozeen vs. sloshy.
- Stave It Off Guy — A guitarist resembling a typical singer-songwriter, appearing in for kids and Crystal Fortress.
- Email space program — The filmstrip includes pictures of some very human looking "hot '60s-looking girls". It also includes pictures of Ted Averill and his co-worker, who appear human.
