In the next email, it reveals who tampered with it. Next, it disappears, followed by someone asking if they've found it. Finally, it appears as if they have either found it, or may find it soon. However, in Email 170, it appears that Homestar is aware of the gag as he sighs and says "DNA Evidence" without enthusiasm. Summing it up would be that they are searching for tampered DNA Evidence.

In six consecutive Strong Bad Emails, characters have referred to DNA evidence. DNA is often used in forensic science deduction to connect bodily fluids or tissue found at a crime scene to a particular individual.
