Metroid is a series of science fiction action/adventure games developed and published by Nintendo. References to the series have been made several times in the Homestar Runner body of work.

- Email invisibility — During an Easter egg, the Tandy 400 references the password "JUSTIN BAILEY" from Metroid for the NES.
- Email japanese cartoon — An Easter egg shows the ending to Metroid for the NES. Additionally, "Maru Maru" is listed in the 20X6 cartoon's credits. It is a misspelling of Maru Mari (丸まり), the Japanese name for the Morph Ball, from Metroid for the NES.
- Main Page 17, Stinkoman 20X6, email alternate universe, and Twenty THANXty Six — The spinning jump that Stinkoman uses resembles Samus's Spin Jump.

- The sound effect for Tampo's destruction is taken from Metroid for the NES.
