Although only approximately 10% of the US population are left-handed, several characters in the Homestar Runner Universe appear to be either left-handed or ambidextrous (even though some characters do not have visible arms).

- Homestar plays the lute left-handed in Tis True, Pom Pom, Tis True.

- Strong Sad plays a classical guitar left-handed in alternate universe.
- Although Strong Bad is right handed, he uses a left-handed marker.
